Titanium dioxide (TiO2) is a multifunctional semiconductor that exists in three crystalline forms: anatase, rutile, and brookite. Owing to an appropriate combination of physical and chemical properties, environmental compatibility, and low production cost, polycrystalline TiO2 has found a large variety of applications and is considered to be a promising material for future technologies. One of the most distinctive physical properties of this material is its high photocatalytic activity (Nam et al., 2019); however, more recently it has attracted growing interest because of its resistive switching abilities (Yang et al., 2008).
In a 2016 study published in Scientifica (Cairo), Egyptian researchers examined the effects of titanium dioxide nanoparticles on the organs of mice by orally administering the food additive daily, for five days. The results showed that the exposure produced “mild to moderate changes in the cytoarchitecture of brain tissue in a time dependent manner.” Furthermore, “Comet assay revealed the apoptotic DNA fragmentation, while PCR-SSCP pattern and direct sequencing showed point mutation of Presenilin 1 gene at exon 5, gene linked to inherited forms of Alzheimer’s disease.” The researchers wrote: “From these findings, “the present study concluded that TiO2NPs is genotoxic and mutagenic to brain tissue which in turn might lead to Alzheimer’s disease incidence.”
Titanium dioxide (E 171) is authorised as a food additive in the EU according to Annex II of Regulation (EC) No 1333/2008.
The inception and evolution of lithopone can be traced back through various industries and diverse applications. Revered for its robust hiding power, this white pigment, also called sulfide of zinc white, has been an invaluable asset to industries requiring a durable and reliable white pigment. Lithopone was an economical and functional solution as an alternative to lead carbonate, which is prone to change, and zinc oxide, known for its brittleness.

JECFA previously assessed titanium dioxide at its 13th meeting, at which time the expert committee assigned a “not specified” ADI for the additive due to an absence of significant absorption and a lack of toxicological effects in the available experimental animal and human studies. Since its original evaluation by JECFA, titanium dioxide has become a public point of contention, with its ban being introduced (and then subsequently withdrawn) in California legislation in 2023, a legal battle playing out in the EU over the additive’s ban and classification as a carcinogen in 2022, and the European Food Safety Authority (EFSA) calling titanium dioxide unsafe. However, supporters of titanium dioxide say that claims about its dangers are founded in unreliable studies, and some recent research has supported its safety as a food additive.
